Just announced on the MythTV developer mailinglist. Starting from Tuesday, December 11, you need to use MySQL 5, if you are running MythTV SVN.
The upgrade should be no problem, since compatibility issues were resolved a long time ago. But of course you should still back up your database before upgrading.
